Heme Dependent Catalase Conditionally Contributes to Oxygen Tolerance of Tetragenococcus halophilus Strains Isolated from Soy Sauce Moromi

چکیده

Tetragenococcus halophilus strains are the halophilic lactic acid bacteria (LAB) that present in microbial communities during soy sauce or other hyperosmotic foods’ fermentation. This species contributes to formation of volatiles fermented foods but may experience harsh conditions such as oxidative stress and osmotic The characterization oxygen tolerance T. elaboration its antioxidant mechanism important for selection suitable LAB food In this work, growth isolated from moromi under both aerobic anaerobic was compared, function their enzymes investigated. These showed differences oxidation resistance, they all produce including superoxide dismutase, peroxidase glutathione reductase. Interestingly, genes encoding catalase (CAT) genome strains, though some them pseudogenes. Catalase produced by belongs heme-dependent CAT, activity could only be detected presence heme condition. CAT conditionally resistance hydrogen peroxide stress. results elucidated possible revealed strains.

Genetic and physiological diversity of Tetragenococcus halophilus strains isolated from sugar- and salt-rich environments.

Tetragenococcus halophilus is known to flourish in extreme salt environments. Recently, this halophilic bacterium also appeared as the dominant microflora during storage of sugar thick juice, an intermediate product of beet sugar production. Although T. halophilus can cause degradation of thick juice, dominance of this bacterium does not always result in degradation. In this study T. halophilus...

Preparation of Phage-insensitive Strains of Tetragenococcus halophila and Its Application for Soy Sauce Fermentation.

We attempted to breed phage-insensitive strains of Tetragenococcus halophila D10. Phage contact during selection initially caused the occurrence of lysogeny. Subsequently, we screened phage-insensitive mutants by replica plating so that mutant cells did not touch the phage during selection. Two strains were selected from about 150,000 strains. They grew normally in soy sauce mash (moromi) in th...

Soy sauce is a salty condiment and is known as shoyu in Japan, chiang-yu (or -yi) in China, kecap in Indonesia, kanjang in Korea, toyo in The Philippines, and see-ieu in Thailand (O’Toole, 1997). In the fermentation of soy sauce in Thailand, Aspergillus oryzae strains have been used for koji production (Lotong, 1998). Zygosaccharomyces rouxii strains were reported to ferment glucose or other su...

Citrate Metabolism by Pediococcus halophilus.

Several strains of non-citrate-metabolizing Pediococcus halophilus have previously been isolated from soy sauce mash or moromi. The factors controlling the metabolism of citrate in soy pediococci were studied. All the soy pediococcal strains tested which failed to decompose citrate did not possess citrate lyase [citrate (pro-3S)-lyase; EC 4.1.3.6] activity. In P. halophilus, citrate lyase was a...

Aspartate Decarboxylation Encoded on the Plasmid in the Soy Sauce Lactic Acid Bacterium, Tetragenococcus halophila D10.

Tetragenococcus halophila D10 decarboxylates aspartate to alanine, but T. halophila D10 derivatives generated by a curing treatment could not (Asd(-) derivatives). We observed by electrophoresis three plasmid bands in T. halophila D10; all Asd(-) derivatives lost the largest of these bands. This plasmid, pD1, has two SalI sites. We cloned and sequenced the 10 kb SalI fragment. The DNA sequence ...
